Yatsushiro castle is a castle in Yatsushiro, Kumamoto prefecture. Throughout the Edo period, it was the seat of the Matsui family, karô to the Hosokawa clan of Kumamoto han.

The first Matsui family lord of Yatsushiro was Matsui (Nagaoka) Okinaga. In 1632, in following with the shogunate's wishes, Hosokawa Tadaoki, lord of Kumamoto, declared that following his death, Matsui would become castellan of Yatsushiro.
