Hotta Masamori served as rôjû from 1633 until his death in 1651. He served on the hyôjôsho, and was named chamberlain in 1640.

Masamori's third son, Hotta Masatoshi, also joined the rôjû and became Tairô later in life. Masamori's fifth son, meanwhile, was adopted by Nanbu Shigenao, coming to be known as Nanbu Katsunao.

Masamori committed junshi seppuku, following his lord Shogun Tokugawa Iemitsu in death.
