Sanesue was the second son of Ando Yoshisue based in Mutsu province. He defeated the Onodera in battle in 1588 and clashed with the Nanbu family. He sided with Tokugawa Ieyasu during the Sekigahara Campaign (1600) but was unable to assist in the field due to an illness. He was transferred by the Tokugawa to Shisido in Hitachi in 1602 and, in 1632, to Asama in Ise province.
